Yes, these memory barriers are added by the compiler when you use volatile variables.

Actually the current design of WROVER board has this very problem with pullup/pulldown resistors you have mentioned. You can find SD card signals in PIN MUX documentation, they have HS2_ prefix.

HS1_ is another SD/MMC slot, with 8 data lines, but it will not be initially supported in software.

Low speed signals (like card detect) can indeed be routed via gpio matrix.

One more note: it is possible to power down RTC fast memory (it just is kept powered on by default). If you do so with the current chip revision, it will resume from deep sleep but will experience a watchdog reset and then proceed to normal boot. This extra time spent waiting for wdt reset may cance...

Wrover-kit would need some pull-up resistors added. Otherwise, yes, it will work.

C standard library does some buffering for you, it reads into a buffer stored in the FILE structure, and then copies data into the buffer you have passed to fread. Now why the data doesn't get read, I cannot say. Can you please add some debugging to see how many bytes SPIFFS_read gives you?

Also IIRC on Windows you may need to press "Boot" button before uploading and hold it until the upload starts.
