I wanted to show how reset and boot signals are generated from CH343 DTR/RTS, but cannot add a picture here because this error
"Sorry, the board attachment quota has been reached." :(

It looks, when cable is disconnected, you are in the state with DTR = 1 and RTS = 0.
Speculating again based on the schematic here

a reason for this state might be the fact that CH343 has no main power - VDD5 (VBUSB in the schematic), but VIO has (connected to 3.3V)
So i suggest you take a cheap USB cable with only 2 wires (+5V and gnd, used only for charging) and power your board and see if its working.
